The road has been closed on the N56 between Termon and Kilmacrennan.
Gardaí in Donegal said the road was closed 'following a serious injury traffic collision.'
A diversion is being set up with the assistance of Donegal County Council.
"We are asking all to be patient," a spokesperson said. "Thank you."
